It's not "cueing up" in the sense that you would with DJ software or a mixer and turntables but if have two sound devices you can set the "Track Editor Audio Device" to be different to the device used for all the other outputs.
If you have (say) a mixer and feed that from a USB sound card you can feed the computer's built in sound output into a different channel and use that for pre-hear.
What PlayIt Live doesn't have (because there is an assumption that complex setups will use an external mixer) is an output which is main mix unless you are using "pre-hear" (also known as Pre Fade Listen - PFL).
